Charrière fut faussement accusé de meurtre en 1931 et condamné à perpétuité au bagne en Guyane française. Le régime là, dit « le chemin de la pourriture », était affreux. Sa récompense fut d'abord un cachot inondé de la mer toutes les marées, puis un séjour à la réclusion, « la mangeuse d'hommes », où il fut puni par la quasi-famine. Mais il désira tant sa liberté que, n'importe combien de ses essais aient échoué, il tenta une évasion après l'autre.
